"Today" show co-host Kathie Lee Gifford, along with her football Hall of Fame legendary husband Frank Gifford, hosted the gala for the 10th consecutive year.
Knight, known as the “Empress of Soul,” sang a number of songs at the Palace Theatre event, which was held May 30.
More than 1,000 people attended the gala, which helps fund research into Niemann-Pick type C disease.
It causes people to be unable to properly metabolize cholesterol and is ultimately fatal; most children with type C die before the age of 10, according to the National Niemann-Pick Disease Foundation. Of those who live longer, most die before the age of 20, with very few reaching age 40.
Andrea and Phil Marella started Dana’s Angels Research Trust in 2002 in hopes of helping their two children who had Niemann-Pick type C.
The Marellas’ daughter, Dana, died two years ago at age 19.
Funding Niemann-Pick research can also potentially help people with other life-threatening issues possibly related to cholesterol, including heart disease and stroke.
